Top-of-the-table UNAM will face fourth-placed Grootfontein in the men's second semi-final, while coastal outfit Kudus will lock horns with Wanderers in the first semi-final on Saturday afternoon.
Wanderers finished second behind UNAM and will face coastal outfit Kudus.
The two teams previously met earlier in the year, with each winning one game at home.
Wanderers won the first game 35-25 at home, while Kudus responded with a 51-27 victory in Walvis Bay.
Kudus head coach Shaun White credited his players for the team's improvement throughout the season and is looking forward to the clash.
White's opponent on the sidelines, Dirk Human, will go into the match without star players lock Johan Meyer and flanker Peter Diergaardt.
In the other semi-final, UNAM will look to continue their impressive season as they face a rejuvenated Grootfontein team, which secured a fourth-place position, three points ahead of defending champions United.
Both teams have been hard at work in training ahead of Saturday and are expecting tough matches.
The afternoon will be preceded by two reserve league matches, with Wanderers 2 facing UNAM 2 and Kudus 2 taking on Rehoboth 2.
